A Study on the HLA Class I Genotyping: (1) HLA-A Alleles



Abstract
Recent advances in the molecular biology of HLA class I genes have revealed HLA class I allelic diversity and polymorphism at the DNA sequence level. The genotyping of HLA-A was performed on the 115 personsly who were previousiy defined the
serotypes,
using the polymerase chain reaction and dot-blot hybridization with allele specific oligonucleotide probe (PCR-ASO). All the HLA class I genes were amplified through the first PCR, and then the first PCR products were used as template DNA for the
second
PCR to amplify the gene between exon 2 and exon 3 of HLA-A. Thirty two ASOs were used to define the HLA-A alleles. We compared the genotypes with the serotypes in each samples. The genotypes of A3, A11, A24, A26 and A30 showed good concordance
with
the
serotypes(0.89subtypes in
A2, A11, A24, A29, A30 and A33 by hybridization patterns. And some novel hybridization patterns were found in A2, A11, A24, A26, A29, A30 and A33. These results suggest that the genotyping of HLA-A can be used to study the subtypes or to define a
novel
alleles in Korean.
